Verenium’s MedEv™ platform enables the optimization of key properties of candidate therapeutic or diagnostic antibodies or proteins through Verenium's suite of DirectEvolution® technologies. Our comprehensive and proprietary platform has proven utility in improving a variety of properties, including affinity, specificity, effector functions, expression levels, stability, and solubility. In addition, our Human Framework Reassembly™ platform provides rapid optimization of non-human antibodies.
The robust gene evolution engine that powers Verenium's MedEv™ platform has been successfully applied to over 40 protein and antibody programs.
Three comprehensive protein engineering techniques comprise the MedEv antibody optimization process: (HuFR™), Human Framework Reassembly, Gene Site Saturation Mutagenesis™ (GSSM™) and Tunable GeneReassembly™ (TGR™) technologies (see diagram).
Verenium’s HuFR™ technology optimizes antibodies by combining a proprietary library of human framework regions with the Complementarity Determining Regions (CDRs) of a parental non-human antibody-generally a murine, or mouse, antibody. Optimized antibodies can be further optimized using Verenium’s GSSM™ approach. This technology creates a library of single point mutations by incorporating all of the 20 amino acids at every position along a protein’s sequence. Our GeneReassembly™ technology is used to combine the best GSSM™ variants into a single gene for the creation of a superior antibody.
